Why does Van Helsing know a High Tier Vampire running a brothel?

Because Van Helsing’s favorite things are Gwent Card and visiting brothels. He was probably a playboy in his youth. As for why the Demon Hunter didn’t start a fight with the vampires, the reason is simple; as Van Helsing said, they are not heroes, just a bunch of experts dealing with monsters.
Before they arrived, Van Helsing ntioned that this brothel is quite special.
The vampires here, in addition to the required monetary paynt, charge a bit of blood as compensation after the service.

She smiled mysteriously and turned to the vampires, saying, "Close up."
